SYNTHESIS OF NEW TRIAZOLO[1,5-A]PYRIDINE DERIVATIVES AND THEIR LIFE SPAN EXTENDING EFFECT ON C. ELEGANS

Abstract:

In the present study the synthesis of new 1,2,4-triazolo[1,5-a]pyridines and their fused ring systems was reported. The chemical structure of the synthetic compounds and the mechanism of their formation were examined. The new synthesized compounds were subjected to various types of activity like antimicrobial, antioxidative, anti-inflammatory activities. The effect of the novel synthesized compounds on the life span of C. elegans as well as their effect on the induced stress resistance in the nematode C. elegans in vivo was studied. Some of these new compounds were found to extend lifespan of wild type C. elegans under standard laboratory conditions. Both heat stress- and induced chemical stress- resistance of the wild type animals were improved in a dose dependant manner due to treatment of these compounds. In addition, treatment of worms with some of these synthesized new compounds was found to significantly attenuate the formation of advanced glycation end products and malondialdehyde in a dose dependent manner. In conclusion, the life span extending effect of these compounds as well as their protective effect against both heat- and chemical-induced stress might be through their antioxidant effect. These findings are supported by the data obtained from the effect of these compounds on the lipid peroxidation and protein glycation. These data demonstrated a potential role of new 1,2,4-triazolo[1,5-a]pyridine derivatives in attenuating oxidative stress markers in the nematode C. elegans.
